Rethinking the Wrinkling: Key Genes Cause Aging

jerry submitted, created time 2 months 1 day (www.sciam.com)

Scientists may have been thinking about the causes of aging all wrong. Instead of being the result of an accumulation of genetic and cellular damage, new evidence suggests that aging may occur when genetic programs for development go awry.

Cell: A key pathway of acute lung injury-Oxidative

kavin submitted, created time 7 months 6 hours (www.cell.com)

Multiple lung pathogens such as SARS cause high lethality due to acute respiratory distress syndrome. The researchers, through experiments in mice, indentify oxidative stress and innate immunity as key lung injury pathways that control the severity of ALI.

Nrf2 mediates cancer protection but not prolongevity induced by caloric restriction

davidd submitted, created time 8 months 4 weeks (www.pnas.org)

Caloric restriction (CR) is the most potent intervention known to both protect against carcinogenesis and extend lifespan in laboratory animals. A variety of anticarcinogens and CR mimetics induce and activate the NF-E2-related factor 2 (Nrf2) pathway. Nrf2, in turn, induces a number of antioxidative and carcinogen-detoxifying enzymes. Thus, Nrf2 offers a promising target for anticarcinogenesis and antiaging interventions. They used Nrf2-disrupted (KO) mice to examine its role on the biological effects of CR

Live slow die young

jane2007 submitted, created time 9 months 3 weeks (www.nature.com)

Active people could be up to 10 years "younger" than couch potatoes, at least according to one measure of biological age.

FOXO and insulin signaling regulate sensitivity of the circadian clock to oxidative stress

bianjie submitted, created time 1 year 1 month (www.pnas.org)

Circadian rhythms can be regulated by many environmental and endogenous factors. We show here a sensitivity of circadian clock function to oxidative stress that is revealed in flies lacking the foxo gene product. When exposed to oxidative stress, wild-type flies showed attenuated clock gene cycling in peripheral tissues, whereas foxo mutants also lost behavioral rhythms driven by the central clock.

Oxidative stress causes bone loss in estrogen-deficient mice through enhanced bone marrow dendritic cell activation

scott submitted, created time 1 year 2 months (www.pnas.org)

Increased production of tumor necrosis factor (TNF) in the bone marrow (BM) in response to both oxidative stress and T cell activation contributes to the bone loss induced by estrogen deficiency, but it is presently unknown whether oxidative stress causes bone loss through T cells.

Faster rates of post-puberty kidney deterioration in males is correlated with elevated oxidative stress in males vs females at early puberty

annatto submitted, created time 1 year 4 months (www.biomedcentral.com)

"This data suggest that early puberty, with gender-related elevation in oxidative stress in males, is a key compromising factor on kidneys in males."

Trimetazidine improves endothelial dysfunction in chronic heart failure: an antioxidant effect

amanda submitted, created time 1 year 6 months (eurheartj.oxfordjournals.org)

"TMZ improves the ED-relaxation in patients with ischaemic cardiomyopathy. The antioxidant properties of TMZ may play a role. "

Paraoxonase-2 Reduces Oxidative Stress in Vascular Cells and Decreases Endoplasmic Reticulum Stress-Induced Caspase Activation

fiona submitted, created time 1 year 7 months (circ.ahajournals.org)

"In the vascular system, elevated levels of reactive oxygen species (ROS) produce oxidative stress and predispose to the development of atherosclerosis. Therefore, it is important to understand the systems producing and those scavenging vascular ROS. Here, we analyzed the ROS-reducing capability of paraoxonase-2 (PON2) in different vascular cells and its involvement in the endoplasmic reticulum stress pathway known as the unfolded protein response."

Genetic deletion of p66Shc adaptor protein prevents hyperglycemia-induced endothelial dysfunction and oxidative stress

cappuccion submitted, created time 1 year 7 months (www.pnas.org)

Increased production of reactive oxygen species (ROS) and loss of endothelial NO bioavailability are key features of vascular disease in diabetes mellitus. The p66Shc adaptor protein controls cellular responses to oxidative stress. Mice lacking p66Shc (p66Shc–/–) have increased resistance to ROS and prolonged life span. The present work was designed to investigate hyperglycemia-associated changes in endothelial function in a model of insulin-dependent diabetes mellitus p66Shc–/– mouse. p66Shc–/– and wild-type (WT) mice were injected with citrate buffer (control) or made diabetic by an i.p

Tropospheric Ozone Toxicity vs. Usefulness of Ozone Therapy

biscuits submitted, created time 1 year 8 months (www.arcmedres.com)

There is a general consensus that continuous inhalation of air polluted with ozone is detrimental for the lungs and vital organs. Even if the concentration of tropospheric ozone is slightly above the tolerated dose, toxicity ensues owing to the cumulative dose inhaled for months. However, in medicine ozone is used as a real drug and a precise concentration and therapeutic dosage must be calibrated against the antioxidant capacity of blood. As ozone reacts with blood, it generates pharmacological messengers such as H2O2 and lipid oxidation products (LOPs)
